I do have an opinion, which is that it’s only one difference, for two reasons.
First, I’ve always believed, and last Saturday, Nighthawks answered a similar question the same way I think of it, that if two changes happen to one object, in this case, Claude’s shirt front, it’s one difference.
Second… his collar doesn’t actually change anyway.
You see a curved line under his chin in panel 1 that could be a seam, ie the edge of a rib knit collar, only partially drawn.
A lot of things in cartoons are partially suggested.
Or, it could be a short wrinkle caused by Claude’s chin or his body shape… or just a little artistic flourish.
In any case, when the jacket is zipped in panel 2, the edges of the jacket cover the sides up to that point.
Does it continue under the jacket where we can’t see it?
We can’t tell, but probably not, as it’s meant to be the same line as in panel 1.

The line you think you see is the edges of the jacket, not a continuation of the collar.
Could you get out your micrometer and find a tiny discrepancy in the length of that middle line?
Probably, but the artist is human, not using precision drafting tools, and we dont count such tiny glitches as differences.
